为探究RNA聚合酶(RNA polymerase)结合与转录过程的关联,本研究以富营养培养基中培养的大肠杆菌(Escherichia coli)为研究对象,在其不同生长阶段下,针对持家型SigmaD因子与RNA聚合酶β亚基开展染色质免疫共沉淀测序(ChIP-seq),并将测序结果与RNA测序(RNA-seq)数据进行整合分析。实验设计概述:通过染色质免疫共沉淀测序联合RNA测序,绘制RNA聚合酶结合位点与转录的相关图谱。
