Multi-process metagenomic analysis is prone to experimental bias, and improving its reliability is an urgent issue. Therefore, it has been proposed to use control samples, called mock communities, which can measure the accuracy of the experimental process, for metagenomic sequencing analysis. Our goal is to develop new mock communities suitable for various microbiome studies, and we focused on a method to measure the amount of genomic DNA in bacterial cells to produce high-quality mock communities. Shotgun sequencing was used to validate the relative abundance of multiple mixed strains. Our results showed that this method is suitable for achieving our goal.
多流程宏基因组分析(metagenomic analysis)易引入实验偏差,提升其可靠性是亟待解决的关键问题。因此,学界提出采用被称为模拟群落(mock community)的对照样本开展宏基因组测序分析,此类样本可用于量化实验流程的准确性。本研究旨在开发适用于各类微生物组研究的新型模拟群落,为此我们聚焦于一种可定量细菌细胞内基因组DNA含量的方法,以制备高质量的模拟群落。我们采用鸟枪测序法(Shotgun sequencing)对多种混合菌株的相对丰度进行验证。研究结果表明,该方法可有效实现本研究的既定目标。
