tRNA-derived small RNAs (tsRNA) are a new type of noncoding RNAs that can be mainly classified into two groups: tRFs (tRNA-derived fragments) and tiRNAs (tRNA halves). The abnormal expression of tsRNAs is known to play an important role in the biological progression of breast cancer. However, it's still unclear about the mechanism of tsRNAs in cancer. tRF-1-Ser is a tRF that is high expression in breast cancer and negatively regulated by 25(OH)D. Our study aims to find out the effect of tRF-1-Ser on breast cancer and explore the change of RNA in tRF-1-Ser knock-down breast cancer cells. Overall design: To explore the influence of tRF-1-Ser on breast cancer, we transfected MDA-MB-453 with shtRF-1-Ser by lentivirus and verified the efficiency of knock-down by qPCR. The groups that MDA-MB-453 transfected with shtRF-1-Ser or shNC are named lv_shtRF or lv_con. Each group was duplicated three times.

实验设计概述:为探究tRF-1-Ser对乳腺癌的调控作用,本研究通过慢病毒将shtRF-1-Ser转染至MDA-MB-453细胞,并采用qPCR验证敲低效率。将转染shtRF-1-Ser与阴性对照shNC的MDA-MB-453细胞组分别命名为lv_shtRF与lv_con,每组设置3次重复。
