We aimed to interrogate the embryoâmaternal interface for embryo-induced changes in the global transcriptome of bovine endometrial epithelial cells (bEECs). In vitro-produced day-5.5 morulae were cultured in juxtacrine apposition (Contact+) with bEECs, or in transwells (Contact?), for 48 h. A group of bEECs cultured in the absence of embryos (Control bEEC) was also included. After co-culture, bEECs were recovered for transcriptome analysis. Overall design: Bovine endometrial epithelial cells (bEECs) were co-cultured in juxtacrine (Contact+) or non-juxtacrine (Contact-) apposition with in vitro-derived morulae for 48 h. bEECs transcriptomes were sequenced (NGS) on the BGISEQ-500 platform.
本研究旨在解析胚胎-母体界面,探究胚胎诱导的牛子宫内膜上皮细胞(bovine endometrial epithelial cells,bEECs)全局转录组变化。将体外制备的第5.5天桑椹胚,分别与bEECs以邻位并置方式(Contact+组)共培养,或置于Transwell小室中(Contact−组)共培养48小时;同时设置未添加胚胎的bEECs培养组作为对照(Control bEEC组)。共培养结束后,回收bEECs用于转录组分析。整体实验设计如下:将牛子宫内膜上皮细胞(bEECs)与体外来源的桑椹胚,分别以邻位接触(Contact+)或非邻位接触(Contact−)的方式共培养48小时,随后采用BGISEQ-500平台对bEECs转录组进行下一代测序(NGS)。
