scRNA-seq of CD45+ leukocytes isolated from WT and triple mutant-infected mice identified several genes with concordant changes in H3Ac status and transcriptional expression. Overall design: Live, CD45+ leukocytes were isolated by FACs from implant-associated tissue at day 14-post infection for scRNA-seq. While these samples were prepared and sequenced for single-cell RNA-seq, they were analyzed as bulk RNA for this study. All raw data for a single-cell analysis have been included.
从野生型(Wild Type, WT)和三重突变体感染小鼠中分离的CD45+白细胞的单细胞RNA测序(scRNA-seq)分析,鉴定出多个组蛋白H3乙酰化(H3Ac)修饰状态与转录表达水平呈现一致性变化的基因。
整体实验设计:于感染后第14天,通过荧光激活细胞分选术(Fluorescence-Activated Cell Sorting, FACS,原文标注为FACs)从植入物相关组织中分离活的CD45+白细胞用于scRNA-seq。尽管本研究已按照单细胞RNA测序的流程完成这些样本的建库与测序,但本次分析将其作为批量RNA样本开展。所有用于单细胞分析的原始数据均已纳入本数据集。
