Epithelial-mesenchymal transition (EMT) is a process by which epithelial cells lose cell-cell contact and gain cancer malignancy such as invasion, stemness, chemoresistance and metastasis. Reverse precess, mesenchymal-epithelial transition (MET) is also important for colonization. Extracellular vesicles (EVs) secreted from cancer cells are also important for cancer malignancy. To analyze RNAs from cells and EVs during EMT and MET, RNA sequencing was performed using E-cadherin-RFP/Py2T reporter system.
上皮间质转化(Epithelial-mesenchymal transition, EMT)是指上皮细胞丧失细胞间连接,并获得侵袭、干细胞性、化疗耐药及转移等恶性表型的生物学过程。而间质上皮转化(Mesenchymal-epithelial transition, MET)这一逆向过程,对肿瘤定植同样具有关键意义。癌细胞分泌的细胞外囊泡(Extracellular vesicles, EVs)同样与肿瘤恶性表型密切相关。为分析上皮间质转化与间质上皮转化过程中细胞及细胞外囊泡内的RNA表达情况,研究人员采用E-钙粘蛋白-红色荧光蛋白(E-cadherin-RFP)/Py2T报告系统开展了RNA测序实验。
