The goal of this study was developed a bioinformatic tool for discovering and selection molecular markers using NGS data.These markers will be applied for phylogenetics, population genetics and phylogeography studies.We sequenced (454 GS-FLX Titanium) intra- and interspecific pooled data from three individuals of the Nemesia genus (two individual of N. raripilia and one individual of Nemesia sp.) and one of the Iberesia genus (Iberesia brauni).RRL was prepared using the enzyme NotI and the size selection of the DNA fragments was performed by excision from an electrophoretic gel.

本研究旨在开发一款基于下一代测序(Next-Generation Sequencing, NGS)数据发掘及筛选分子标记的生物信息学工具。该类标记将应用于系统发育学、群体遗传学及系统地理学相关研究。我们采用454 GS-FLX Titanium测序平台,对沟酸浆属(Nemesia)的3个个体(2个为N. raripilia,1个为未定名沟酸浆种Nemesia sp.)以及伊比利亚属(Iberesia)的1个个体(Iberesia brauni)的种内与种间混合测序样本开展了测序工作。本研究使用限制性内切酶NotI制备了RRL文库,并通过从电泳凝胶中切胶的方式完成了DNA片段的大小筛选。
