Rat mast cell line RBL-2H3 was analyzed to investigate the molecular mechanism of Dectin-1-mediated activation and responses of mast cells. Dectin-1-mediated signaling stimulated characteristic gene expression, such as MCP-1, IL-4, IL-13, TNF-αand Nfkbiz. Overall design: RBL-2H3 cell line stably expressing Dectin-1 was established. Non-stimulated cells (control) compared to Dectin-1-stimulated (Curdlan) cells in order to comfirm Dectin-1-mediated signaling-inducible genes. Dectin-1-stimulated cells compared to Syk inhibitor R406-pretreated cells in order to examine the importance of Syk for Dectin-1-mediated gene up-regulations.
本研究以大鼠肥大细胞系RBL-2H3为研究对象,旨在探究Dectin-1(Dectin-1)介导的肥大细胞活化与应答的分子机制。Dectin-1介导的信号通路可诱导特征性基因表达谱,涉及单核细胞趋化蛋白1(MCP-1)、白细胞介素4(IL-4)、白细胞介素13(IL-13)、肿瘤坏死因子α(TNF-α)及Nfkbiz等基因。
实验整体设计如下:首先构建稳定表达Dectin-1的RBL-2H3细胞系。其一,将未受刺激的细胞(对照组)与经凝结多糖(Curdlan)刺激的Dectin-1活化细胞进行比较,以确认Dectin-1介导的信号通路诱导的差异表达基因;其二,将经脾酪氨酸激酶(Syk)抑制剂R406预处理的细胞与Dectin-1刺激后的细胞进行比较,以验证Syk在Dectin-1介导的基因上调过程中的重要作用。
