Xanthocillin resistant mutants in Acinetobacter baumannii

Acinetobacter baumanii reference strain wild-type DSM30007 sensitive to Xanthocillin was exposed to growing concentrations of this antibiotic in 11 rounds of culturing, resulting in selected mutants exhibiting resistance. Shotgun genomic sequencing were obtained from wild type strain, from resistant isolates at different stages of the 11 rounds of increasing selection, from strains with the same amount of generations without antibiotic selection representing accumulated random genetic drift. The objective was to find the gene mutations responsible for the selected resistance.

对黄青霉素（Xanthocillin）敏感的鲍曼不动杆菌（Acinetobacter baumanii）野生型参考菌株DSM30007，被置于浓度梯度递增的该抗生素环境中开展11轮传代培养，最终筛选获得携带耐药表型的突变株。研究团队分别对野生型菌株、11轮递增筛选不同阶段的耐药分离株，以及经过相同传代代数且未施加抗生素选择压力、仅发生随机遗传漂变的菌株开展鸟枪基因组测序（Shotgun genomic sequencing）。本研究的目标为筛选鉴定与该耐药性产生相关的基因突变。
