Vertical transmission of Trypanosoma cruzi (T. cruzi) has become a globalized health problem. The main mechanisms that lead to fetal infection by this parasite remain poorly understood. To model the human placenta environment, we used a three-dimensional (3D) cell culture system of syncytiotrophoblasts (SYNs) and showed that 3D-grown SYNs are highly resistant to T. cruzi infection. We explored the immunological signatures that drive SYNs infection control using RNA sequencing and whole transcriptome analysis.

克氏锥虫(Trypanosoma cruzi,T. cruzi)的垂直传播已成为一项全球化的公共卫生难题。目前,该寄生虫引发胎儿感染的核心机制仍未明确。为模拟人类胎盘环境,我们构建了合体滋养层细胞(syncytiotrophoblasts,SYNs)的三维(3D)细胞培养体系,实验显示三维培养的合体滋养层细胞对克氏锥虫感染具有极强的抵抗力。我们通过RNA测序(RNA sequencing)与全转录组分析(whole transcriptome analysis),探究了调控合体滋养层细胞感染抑制的免疫特征。
