We examined the ability of Gal-9 to inhibit cell proliferation via apoptosis, and the effects of Gal-9 on cell cycle-related molecules in various human colon and colorectal cancer cell lines. In addition, Gal-9-mediated changes of activated tyrosine kinase receptors and angiogenic molecules were assessed using protein array chips in colon and colorectal cancer cells. Moreover, miRNA array analysis was performed to examine Gal-9-induced miRNA expression profiles. We also elucidated if Gal-9 inhibited tumor growth murine in vivo model. In the study, effect of galectin-9 was examined using colon cancer cell lines and xenograft animal model.

本研究首先探究了半乳糖凝集素9(Gal-9)通过细胞凋亡抑制细胞增殖的能力,以及其对多种人结肠及结直肠癌细胞系中细胞周期相关分子的调控效应。此外,本研究借助蛋白芯片(protein array chips)技术,分析了结直肠癌细胞中Gal-9介导的活化酪氨酸激酶受体与血管生成相关分子的表达变化。进一步,本研究通过miRNA芯片分析(miRNA array analysis),考察了Gal-9诱导的微小RNA表达谱改变。同时,我们还在小鼠体内模型中验证了Gal-9对肿瘤生长的抑制作用。本研究采用结肠癌细胞系与异种移植动物模型,对Gal-9的生物学功能进行了系统性考察。
