This project sought to use the latest in DNA sequencing technologies to assemble sequence from Trifolium pratense (red clover) into continuous, chromosome-scale scaffolds. The assembly was generated by deep sequencing using PacBio HiFi reads, and scaffolding was accomplished using Omni-C linkage data. Finally, linkage-group scaffolds were created using a combination of EST linkage map data and Oxford Nanopore reads. The integration of many different technologies and orthogonal datasets resulted in the most continuous assembly of red clover yet released.

本研究旨在借助最新的DNA测序技术,将红三叶草(Trifolium pratense)的基因组序列组装为连续的染色体水平支架(scaffold)。该组装通过PacBio HiFi读长的深度测序生成,支架构建环节借助Omni-C连接数据完成。最终,结合表达序列标签(Expressed Sequence Tag)连锁图谱数据与牛津纳米孔(Oxford Nanopore)测序读长,构建了连锁群支架。多种技术与正交数据集的整合,得到了目前已发布的连续性最优的红三叶草基因组组装结果。
