本发明属于生物技术领域,尤其是植物组织培养技术领域,公开了一种同时去除植物组织培养时外植体表面和内生菌的方法。本发明采用50%的多菌灵对扦插土壤进行消毒处理,采用含有200毫克每升两性霉素B和100毫克每升硫酸链霉素的无菌水溶液对扦插枝条进行消毒处理,采用含有100毫克每升两性霉素B和50毫克每升硫酸链霉素的无菌水溶液对半木质化的新发枝条进行预处理,并且采用溶脂混合液对外植体进行预处理,从而能够有效清除杨树等外植体表面及内部的细菌及真菌,使外植体保持充足的活力。

This invention belongs to the field of biotechnology, particularly the technical field of plant tissue culture, and discloses a method for simultaneously removing surface and endophytic bacteria and fungi from explants during plant tissue culture. This invention uses 50% carbendazim to disinfect cutting soil, disinfects cutting branches with a sterile aqueous solution containing 200 mg/L amphotericin B and 100 mg/L streptomycin sulfate, preprocesses semi-lignified new shoots with a sterile aqueous solution containing 100 mg/L amphotericin B and 50 mg/L streptomycin sulfate, and uses a lipid-dissolving mixture to preprocess explants. This approach can effectively eliminate bacteria and fungi on the surface and inside of explants such as poplars, while maintaining sufficient viability of the explants.
