At 35 DAP whole kernels (pericarp + endosperm + embryo) without glumes of green house grown ears of heterozygous (+/bt2-H2328), self-pollinated plants were visually divided into pools of phenotypically normal looking kernels (small indentation, slightly smaller than mutant kernels, genotype +/+ or +/bt2-H2328) and pools of phenotypically mutant kernels (plump, round kernels, slightly larger than normal kernels, genotype bt2-H2328/bt2-H2328). Pools consisted of 4 kernels. 3 different ears were used for a biological duplicate. Keywords: gene knock out Overall design: 4 arrays - maize
授粉后35天(Days After Pollination, DAP)时,采集杂合子(+/bt2-H2328)自交植株的温室种植果穗,分离得到去除颖壳(glumes)的完整籽粒,即果皮(pericarp)+胚乳(endosperm)+胚(embryo)。通过肉眼表型观察,将籽粒分为两组混样:表型正常籽粒组(籽粒具轻微凹陷,略小于突变体籽粒,基因型为+/+或+/bt2-H2328)与表型突变籽粒组(籽粒饱满圆润,略大于正常籽粒,基因型为bt2-H2328/bt2-H2328)。每组混样包含4个籽粒,共使用3个不同果穗完成生物学重复。关键词:基因敲除(gene knock out)。实验整体设计:共设置4张芯片,实验材料为玉米(maize)。
