An intricate interplay among Tcf1/Lef1, CTCF and Stat5 that orchestrates genomic architecture to coordinately promote CD8+ T cell homeostatic proliferation Overall design: Splenocytes were harvested from hCD2-Cre, Rosa26-lsl-GFP, Tcf1/Lef1 wild-type mice, and sorted for GFP+TCRbeta+ naïve CD8+ T cells. The live cells wer used for CTCF ChIP-seq.
Tcf1/Lef1、CTCF与Stat5之间存在复杂的相互调控网络,三者协同塑造基因组三维结构,共同促进CD8+ T细胞的稳态增殖。整体实验设计:从携带hCD2-Cre与Rosa26-lsl-GFP的Tcf1/Lef1野生型小鼠中分离脾脏淋巴细胞,分选获得GFP+TCRβ+初始CD8+ T细胞,取其中活细胞用于CTCF染色质免疫沉淀测序(ChIP-seq)。
