RNA sequencing of sorted cell types, isolated from mice kidneys, after unilateral ureteral obstructive (UUO) surgery after 2 days, 7 days and a reversible (rUUO) 14 day model with corresponding sham operated mice. The goal of the study was to identify temporal gene expression changes in different cell types corresponding with kidney injury, in order to understand cell specific transcriptomic changes in the context of kidney injury and repair. Overall design: Each group was sequenced from 4 mice, and 4 cell types. 2 day UUO kidneys were sorted into PDGFRβ+, F480+, LTL+ and CD31+ cell types before the RNA was extracted and small RNA sequenced for 4 mice. The same protocol was followed for 7 day UUO mice, rUUO mice, as well as sham operated mice for reference.
本数据集包含从单侧输尿管梗阻(unilateral ureteral obstructive, UUO)术后2天、7天的小鼠肾脏,以及可逆性单侧输尿管梗阻(reversible UUO, rUUO)14天模型小鼠的肾脏中分离分选得到的细胞类型的RNA测序数据,同时设置对应假手术组小鼠作为对照。本研究旨在鉴定与肾损伤相关的不同细胞类型的时序性基因表达变化,以解析肾损伤与修复过程中细胞特异性的转录组改变。
总体实验设计:每个分组均使用4只小鼠的4种细胞类型进行测序。针对4只UUO术后2天的小鼠,先将其肾脏组织分选为PDGFRβ+、F480+、LTL+及CD31+四种细胞类型,随后提取RNA并开展小RNA测序。7天UUO小鼠、rUUO小鼠及作为参照的假手术小鼠均采用相同实验流程。
