Thermoanaerobacter sp. X514 was grown on 1, 10, 50 and 200 mM xylose in order to identify pathways and transporters responsible for xylose transport and conversion to ethanol. Overall design: In this study, X514 cells were grown in difeined medium supplemented with 1, 10, 50 or 200 mM xylose as the sole carbon substrate. Each sample was harvested at mid-log phase. Each sample was run in triplicate using genomic DNA as the control.
为鉴定介导木糖转运及乙醇生成的代谢通路与转运蛋白,本研究以1 mM、10 mM、50 mM及200 mM木糖为唯一碳源,培养热厌氧杆菌属(Thermoanaerobacter sp.)菌株X514。
整体实验设计:本研究中,X514菌株在化学成分确定培养基(defined medium)中培养,以1 mM、10 mM、50 mM或200 mM木糖作为唯一碳源底物。所有样本均于对数中期收获,每组均以基因组DNA作为对照并设置三次生物学重复。
