Advanced glycation end products promote the metastasis of colorectal cancer cells via centrosome amplification mediated by KLF5-CEP57L1 axis

Emerging evidence suggests that diabetes and centrosome amplification (CA) are associated with metastasis and in vitro invasion of cancer cells, respectively. We have reported that diabetes promotes CA; thus, we sought to determine whether CA may serve as a biological link between diabetes and metastasis. Our study demonstrated that advanced glycation end products (AGEs) in vitro simultaneously promoted CA, migration and invasion of HCT116 colorectal cancer cells, with higher levels of CA in migrated cells. Herein, we provided the original western blot and transcriptomic profling results.
新兴研究证据表明,糖尿病与中心体扩增(centrosome amplification, CA)分别与癌细胞转移及体外侵袭存在关联。本团队此前已有研究证实糖尿病可促进中心体扩增,因此本研究旨在探究中心体扩增是否可作为糖尿病与癌症转移之间的生物学纽带。本研究结果显示,体外环境下的晚期糖基化终末产物(advanced glycation end products, AGEs)可同时促进HCT116结直肠癌细胞的中心体扩增、迁移与侵袭,且迁移细胞中的中心体扩增水平更高。本文附原始蛋白质免疫印迹(Western Blot)及转录组分析结果。
