In the present study we analyzed the effect of cellular senescence on the transcriptome of human mesenchymal stem cells (hMSC; alternatively named mesenchymal stromal cells) from bone marrow. Human MSC were isolated from femoral heads of non-osteoporotic donors after total hip arthroplasty. Overall design: Cells were isolated from human bone marrow according to the previously described protocol (Noth et al., 2002, J Orthop Res, 20/5, 1060-1069) under agreement of the local Ethics Committee of the University of Würzburg. Bone marrow was obtained of femoral heads after total hip arthroplasty due to osteoarthritis and/or hip dysplasia. MSC were replated after reaching 70-90% confluence until they entered state of cellular senescence. RNA samples of control cells were taken from passage 1 or passage 2.
本研究分析了细胞衰老(cellular senescence)对人骨髓间充质干细胞(human mesenchymal stem cells,hMSC,又称间充质基质细胞)转录组的影响。本研究的人骨髓间充质干细胞取自非骨质疏松供体全髋关节置换术后的股骨头。
总体实验设计:按照既往发表的方案(Noth等,2002,《骨与关节研究杂志》,20卷第5期,1060-1069页)分离人骨髓细胞,实验已获得维尔茨堡大学当地伦理委员会批准。骨髓样本取自因骨关节炎和/或髋关节发育不良行全髋关节置换术的股骨头。间充质干细胞在汇合度达到70%~90%时进行传代培养,直至其进入细胞衰老状态。对照组细胞的RNA样本取自第1代或第2代。
