We performed a single-cell transcriptome analysis of colon monocytes and macrophages in 2 mice fed with control diet (chow) and 2 mice fed with high-fat diet for 1 week. CD11b+nonDCs colonic macrophages (liveCD45+Lin-CD11b+CD24- and pos for CD64 or Ly6C) were sorted for each replicate mouse, and used for single cell RNA-seq on a 10x genomics Chromium platform.

本研究对2只饲喂正常对照饲料(chow)以及2只饲喂高脂饲料1周的小鼠的结肠单核细胞与巨噬细胞开展了单细胞转录组分析(single-cell transcriptome analysis)。我们从每只生物学重复小鼠中分选得到结肠CD11b阳性非树突状巨噬细胞(CD11b+nonDCs macrophages),其细胞表型为liveCD45阳性、Lin阴性、CD11b阳性、CD24阴性,且CD64或Ly6C阳性;随后使用10x Genomics Chromium平台对上述分选细胞进行单细胞RNA测序(single cell RNA-seq)。
