Single-Cell RNAseq of PBMCs isolated from heavy drinkers and controls

PBMCs were isolated from heavy drinkers (AUDIT>16) and healthy controls. Patients include both lean (BMI<25) and overweight/obese (BMI>25). PBMCs were ex vivo challenged with PolyI:C (1.5ug/ml for 2 hours) or LPS (10ng/ml for 1 hour). Single-cell RNA-seq was then performed

本研究从酒精使用障碍识别测试（Alcohol Use Disorders Identification Test, AUDIT）得分大于16的重度饮酒者与健康对照个体中分离外周血单个核细胞（Peripheral Blood Mononuclear Cells, PBMCs）。受试对象涵盖体质量偏低（体重指数Body Mass Index, BMI<25）与超重/肥胖（BMI>25）两类人群。将分离得到的PBMCs经体外分别以聚肌胞苷酸（Polyinosinic-polycytidylic acid, PolyI:C，1.5μg/ml 刺激2小时）或脂多糖（Lipopolysaccharide, LPS，10ng/ml 刺激1小时）处理。随后对样本开展单细胞RNA测序（single-cell RNA-seq）实验。
