In this study, adaptive laboratory evolution (ALE) was performed on the wild-type Pichia pastoris GS115 strain, yielding the acid-tolerant strain GSAR capable of withstanding pH 2.6. The key genetic elements responsible for the acid tolerance of GSAR were identified, and the GSAR strain was further applied to the neutralizer-free fermentation for the production of organic acids and full-length recombinant proteins.
本研究以野生型巴斯德毕赤酵母(Pichia pastoris)GS115菌株为出发菌株,通过适应性实验室进化(adaptive laboratory evolution, ALE)技术获得了可耐受pH 2.6环境的耐酸菌株GSAR。本研究鉴定了GSAR耐酸性的关键遗传元件,并将该菌株进一步应用于无中和剂发酵工艺,用于生产有机酸及全长重组蛋白。
