Differential gene expression in response to juvenile hormone analog treatment in the damp-wood termite Hodotermopsis sjostedti (Isoptera, Termopsidae)

Normalized cDNA library was constructed from mRNA that was extracted from various castes/developmental stages of Hodotermopsis sjoestedti collected from Yaku-shima Island, Kagoshima, Japan. We employed 454 FLX Titanium system for sequencing the normalized cDNA library, assembled 454 reads with Newbler v2.5.3, and created a EST contig set. To identify genes that respond to a juvenile hormone analog (JHA) application in pseudergates of H. sjoestedti, we carried out JHA treatments on pseudoergates and Suppressive Subtractive Hybridization (SSH). We determined nucleic acid sequences of gene fragments generated from the SSH, and performed similarity searches for those fragments in the 454 EST contig set, using the blastx and tblastx algorithms. This BioProject entry includes the EST contigs that were obtained from the blast searches.
本研究从采集自日本鹿儿岛县屋久岛的 Hodotermopsis sjoestedti 白蚁不同品级及发育阶段的样本中提取mRNA,进而构建了标准化互补DNA(complementary DNA, cDNA)文库。本研究采用454 FLX Titanium测序系统对该标准化cDNA文库进行测序,使用Newbler v2.5.3软件对454测序读段进行组装,得到表达序列标签(Expressed Sequence Tag, EST)重叠群集。为鉴定该白蚁拟工蚁中响应保幼激素类似物(juvenile hormone analog, JHA)处理的基因,本研究对拟工蚁实施JHA处理,并开展了抑制性消减杂交(Suppressive Subtractive Hybridization, SSH)实验。本研究对SSH实验获得的基因片段进行了核酸序列测定,并基于blastx与tblastx算法,在上述454测序得到的EST重叠群集中对这些片段进行了相似性比对搜索。本次提交的生物项目(BioProject)条目包含了通过上述BLAST搜索得到的EST重叠群序列。
