We use in-depth transcriptome sequencing to design probes for exome capture in 16 Swiss stone pine (Pinus cembra), a conifer with an estimated genome size of 29.3 Gbp and no 17 reference genome available. The final transcriptome consisted of 40,468 transcripts representing 27,979 TRINITY genes, totaling to 69.4 Mbp of sequence. Average and median transcript length were 1711 and 1422 bp, respectively (Fig. S1). N50 was 2380 bp and GC content 42.2%. More than 80% of the contigs (34,010) could be annotated.

本研究采用深度转录组测序技术,为16株瑞士石松(Pinus cembra)的外显子组捕获设计探针。该物种为针叶树,预估基因组大小为29.3 Gbp,暂无可用参考基因组。最终组装得到的转录组包含40468条转录本,对应27979个Trinity(TRINITY)基因,总序列长度达69.4 Mbp。转录本的平均长度与中位长度分别为1711 bp和1422 bp(图S1),N50长度为2380 bp,GC含量为42.2%。超过80%的重叠群(共34010条)可被注释。
