Drug resistance remains a main obstacle for cancer treatment. To reveal differentially expressed genes in chemoresistant cells, we established two pairs of resistant/parental esophageal squamous cell carcinoma (ESCC) cell lines, and performed whole transcriptome sequencing to identify both coding and noncoding RNAs that are significantly changed in chemoresistant cells. We identidied several important genes that contribute to the resistance of ESCC cells to chemotherapies. Overall design: Whole transcriptome sequencing of KYSE450/DDP, KYSE450, YES2/DDP and YES2 cell lines. Two replicates for each arm.

耐药性仍是癌症治疗的主要障碍。为揭示化疗耐药细胞中的差异表达基因,我们构建了两对食管鳞状细胞癌(esophageal squamous cell carcinoma, ESCC)耐药/亲本细胞系,并通过全转录组测序鉴定出化疗耐药细胞中发生显著表达变化的编码RNA与非编码RNA。本研究鉴定出数个可促进ESCC细胞对化疗产生耐药性的关键基因。实验设计概况:对KYSE450/DDP、KYSE450、YES2/DDP及YES2细胞系开展全转录组测序,每组设置两个生物学重复。
