The pathological process of traumatic brain injury (TBI) is complicated, and endothelial cells (ECs) is critical in these pathological and physiological processes. In this study, ECs from surgically removed injury tissues were isolated, and then subjected to single-cell RNAseq to explore their transcriptomic profiles. We profiled isolated cells from injury center and injury periphery tissues of three TBI patients. CD45-CD31+ cell suspensions were generated using magnetic-activated cell sorting (MACS), which were then subjected to scRNA-seq performing a 10X Genomics protocol.

创伤性脑损伤(traumatic brain injury, TBI)的病理过程复杂,内皮细胞(endothelial cells, ECs)在其病理生理过程中发挥关键作用。本研究从手术切除的损伤组织中分离内皮细胞,通过单细胞RNA测序(single-cell RNAseq)探究其转录组特征。我们对3例TBI患者的损伤中心区及损伤周边区的分离细胞进行了转录组分析。通过磁激活细胞分选(magnetic-activated cell sorting, MACS)制备CD45-CD31+细胞悬液,随后采用10X Genomics测序流程完成单细胞RNA测序(scRNA-seq)。
