Trascriptome profiles in the cells of A. aceti wild-type strain and its isogenic aceA glcB mutant during growth in medium contaning ethanol and glucose was determined by NimbleGen Eukaryotic Expression array (4x72K). Overall design: Acetobacter aceti NBRC14818 and its isogenic aceA glcB mutant were cultivated in medium containing ethanol and glucose in Erlenmeyer flask with rotary shaking. Total RNA was extracted when optical density at 600 nm was 0.3. The experiment was performed in duplicate independent cultures.
本研究采用NimbleGen真核表达芯片(4×72K),测定了醋酸杆菌(Acetobacter aceti,A. aceti)野生型菌株及其同基因aceA glcB突变体在含乙醇与葡萄糖的培养基中生长时的细胞转录组谱。
实验设计概述:将醋酸杆菌Acetobacter aceti NBRC14818及其同基因aceA glcB突变体接种于装有含乙醇与葡萄糖培养基的锥形瓶中,进行旋转振荡培养。当培养物的600 nm光密度值为0.3时,提取总RNA。本实验设置两组独立重复培养。
