Effects of sequential administration of phage cocktail, ciprofloxacin, and caspofungin on Staphylococcus aureus and Candida albicans dual species biofilms

Polymicrobial biofilms, are another threat in the post-antibiotic era. Staphylococcus aureus and Candida albicans often occur together, causing infections that are difficult to treat. Therefore, many alternative solutions are investigated, including phage antibiotic synergy (PAS), which may be more effective because of different mechanisms of action. In this work, we investigated how different sequences of administration phage cocktail, ciprofloxacin, and caspofungin will eradicate S. aureus/ C. albicans in liquid culture and biofilm. In liquid culture, the sequence of phages then caspofungin then ciprofloxacin completely eradicated S. aureus and C. albicans. In the case of biofilm, the best therapeutic option was the administration of three agents at once or using phages and ciprofloxacin before caspofungin. In conclusion, phages must first be administrated alone or with other factors in both environments. Sequences, when both drugs or only caspofungin was first introduced to cultures, were less effective. The sequence of administration of drugs and phages should be taken into account when planning alternative therapies based on their synergistic action. 多微生物生物膜(Polymicrobial biofilms)是后抗生素时代的又一严峻威胁。金黄色葡萄球菌(Staphylococcus aureus)与白色念珠菌(Candida albicans)常共同定植,引发难治性感染。因此,诸多替代治疗方案正被广泛研究,其中噬菌体抗生素协同作用(phage antibiotic synergy, PAS)因作用机制迥异,或可展现更优疗效。本研究探讨了噬菌体鸡尾酒、环丙沙星(ciprofloxacin)与卡泊芬净(caspofungin)的不同给药顺序,对液体培养体系及生物膜中金黄色葡萄球菌/白色念珠菌的清除效果。在液体培养体系中,先施用噬菌体、再给予卡泊芬净、最后使用环丙沙星的给药顺序,可完全清除目标菌株。针对生物膜体系,最优治疗方案为同时施用三种药剂,或是先给予噬菌体与环丙沙星、后使用卡泊芬净。综上,在两种培养体系中,噬菌体均需率先单独施用,或与其他药剂联合施用。若先施用两种抗菌药物,或仅先给予卡泊芬净,则治疗效果欠佳。基于协同作用开发替代疗法时,应充分考量药物与噬菌体的给药顺序。本数据集附带多个自述文件,可查阅更多相关细节。
