To determine the global gene transcriptional changes in channel catfish anterior kidney after immersion vaccination with attenuated Edwardsiella ictaluri compared to sham vaccinated catfsih control. The attenuated Edwardsiella ictaluri used for vaccination was Aquavac-ESC which was commercially available. Overall design: A six chip study using total RNA samples including 3 control and 3 vaccinated samples. Each RNA sample used for microarray was poold fish anterior kidney samples (5 fish anterior kidney samples were pooled as one). Each chip measrues the expression level of 65,182 genes. For each sequence, 3 probes were selected. Two copies were prepared on arrays.

本研究旨在探究相较于假免疫对照组的斑点叉尾鮰(channel catfish),经减毒迟钝爱德华氏菌(Edwardsiella ictaluri)浸泡免疫后,其前肾组织内的全基因转录组变化。本研究使用的免疫用减毒迟钝爱德华氏菌为市售商品Aquavac-ESC。 实验设计:本研究采用6张基因芯片(microarray)的检测方案,共使用6份总RNA(total RNA)样本,其中3份为对照组样本,3份为免疫组样本。每份用于基因芯片检测的RNA样本均由5条斑点叉尾鮰的前肾组织样本混合制备而成。每张芯片可检测65182个基因的表达水平;针对每条靶序列,共设计3条探针,并在芯片上制备2份重复探针。
