Rice gene global expression analysis upon inoculation with different Magnaporthe isolates

Magnaporthe oryzae is the causative agent of the rice blast, the most relevant rice disease worldwide. To date expression analysis on rice infected with Magnaporthe oryzae have been carried out only with the strains FR13 (leaf) and Guy 11 (root). However different strains of Magnaporthe are present in the environment leading to different rice responses at molecular level. To gain more insight on the unknown molecular mechanisms activated by different Magnaporthe strains during rice defense, a global expression analysis was performed by using the GeneChip® Rice Genome Array. To identify rice genes differentially regulated upon infection by Magnaporthe isolates, inoculation with different strains were performed and samples were collected 24 hours post infection. RNA were obtained from leaf samples after inoculation of rice 2 week-old plantlets with the following strains: rice isolates Magnaporthe oryzae FR13 and CL367, non-adapted strain BR32, isolated from wheat, and Magnaporthe grisea BR29 isolated from crabgrass. Treated and control (mock) rice leaves (cv. Nipponbare) were collected 24 hours post inoculation. Three biological replicates for each interaction type and the corresponding mock were extracted and analysed independently with the GeneChip® Rice Genome Array.

稻瘟病菌(Magnaporthe oryzae)是全球危害最为严重的水稻病害——稻瘟病的病原菌。迄今为止,针对稻瘟病菌侵染水稻的表达谱分析,仅采用了FR13(叶片侵染)与Guy 11(根部侵染)两个菌株开展。然而环境中存在多种稻瘟病菌菌株,会在分子层面引发水稻产生不同的应答反应。为更深入解析不同稻瘟病菌菌株在水稻防御过程中激活的未知分子机制,本研究采用GeneChip®水稻基因组芯片开展了全基因组表达谱分析。为鉴定稻瘟病菌分离物侵染后差异调控的水稻基因,研究团队对不同菌株接种后的水稻样本于侵染后24小时进行采集。本研究以2周龄的水稻幼苗为材料,接种以下菌株后采集叶片样本并提取RNA:水稻来源的稻瘟病菌分离物FR13与CL367、从小麦中分离的非寄主适应性菌株BR32,以及从马唐草中分离的灰梨孢(Magnaporthe grisea)BR29。接种后24小时采集处理组与对照组(Mock,即空白接种对照)的水稻叶片,品种为日本晴(cv. Nipponbare)。每种互作类型及其对应的对照组均设置3次生物学重复,独立采用GeneChip®水稻基因组芯片进行表达谱分析。
