Two syngeneic mouse melanoma cell lines were derived from a cutaneous metastasis (CM) or lymph node (LN) metastasis spontaneously developing in the Mt-Ret transgenic mouse melanoma model. In order to characterize the in vivo growth properties upon inoculation into the skin of syngeneic mice and molecular phenotypes in vivo, we isolated total RNA from established CM and LN melanomas and performed 3’mRNA-Seq analysis for comparison. Gene set enrichment analysis revealed that CM melanomas induced higher interferon pathway activation in the tumor microenvironment than LN melanomas.

两株同基因小鼠黑色素瘤细胞系,源自Mt-Ret转基因小鼠黑色素瘤模型中自发形成的皮肤转移瘤(cutaneous metastasis, CM)与淋巴结转移瘤(lymph node metastasis, LN)。为表征该细胞系接种至同基因小鼠皮下后的体内生长特性与体内分子表型,研究团队从已构建的CM及LN黑色素瘤样本中分离总RNA,并开展3'端mRNA测序(3’mRNA-Seq)分析以进行对比。基因集富集分析结果显示,相较于LN黑色素瘤,CM黑色素瘤可在肿瘤微环境中诱导更为显著的干扰素通路激活。
