不同节段脊髓中存在行使不同功能的神经元。然而,来自不同节段的人源脊髓神经干细胞(human spinal cord-derived neural stem cells, hscNSCs)是否具有节段特性,以及它们在修复脊髓损伤中是否具有差异尚不清楚。在本研究中,我们建立了一个可以高效扩增来自不同发育阶段和节段脊髓组织来源的hscNSCs的培养体系。尽管该培养体系下的细胞都具有神经干细胞特征,但是不同节段来源的hscNSCs在转录组和分化潜能方面都表现出不同的特性。与颈段和腰段来源的hscNSCs相比,胸段来源的hscNSCs对于胸腔脊髓损伤大鼠的修复效果更好。胸段来源的hscNSCs保留胸段胚胎脊髓的特性有助于更好的修复脊髓损伤。该课题强调了神经干细胞节段来源与脊髓损伤部位相匹配的治疗方法,使干细胞移植发挥更好的治疗效果。
Neurons performing diverse functions are distributed across distinct segments of the spinal cord. However, it remains unclear whether human spinal cord-derived neural stem cells (hscNSCs) from different spinal segments possess segment-specific characteristics and exhibit differential efficacy in spinal cord injury repair. In this study, we established a highly efficient culture system for expanding hscNSCs derived from spinal cord tissues at various developmental stages and from different segments. While all cells cultured in this system display typical neural stem cell features, hscNSCs from different segments exhibit distinct transcriptomic profiles and differentiation potentials. Compared to hscNSCs originating from cervical and lumbar segments, thoracic segment-derived hscNSCs show significantly better repair outcomes in a rat model of thoracic spinal cord injury. The retention of characteristics inherent to thoracic embryonic spinal cord by thoracic segment-derived hscNSCs contributes to their enhanced repair capacity for spinal cord injury. This work underscores the therapeutic strategy of matching the segmental origin of neural stem cells with the injured spinal cord site, thereby maximizing the therapeutic efficacy of stem cell transplantation.
