To further decipher the cell fate continuum during reprogramming of T cells to ITNKs, we investigated the gene expression dynamics of T and ITNK cells from a CB sample at single-cell level using micro-well based scRNA-seq technology at different time points post BCL11B ablation T cells that were electroplated with PX458-BCL11B at day 0 (2263 cells), day 5 (1565 cells), day 10 (498 cells), day 15 (204 cells), and day 20 (418 cells), based on sc-RNA seq analysis.
为进一步解析T细胞重编程为固有样自然杀伤T细胞(ITNKs)过程中的细胞命运连续体,我们采用基于微孔的单细胞RNA测序(scRNA-seq)技术,对取自一份脐带血(CB)样本、经PX458-BCL11B电转染以敲除BCL11B的T细胞,在电转染后第0天(2263个细胞)、第5天(1565个细胞)、第10天(498个细胞)、第15天(204个细胞)及第20天(418个细胞)的单细胞基因表达动态开展分析,所有分析均基于单细胞RNA测序结果。
