Through genome skimming for Puya raimondii, P. macrura and P. hutchinsonii, 70 pairs of cross-species SSR primers were selected and synthesized. 42 of 70 pairs of primers were successfully cross-species amplified. After preliminary screening, 12 SSR primer pairs produced clear bands and were used for the detection of potential polymorphisms in 84 Puya raimondii individuals collected from 4 populations in Peru. The number of alleles per locus ranged from one to 6, and the observed and expected levels of heterozygosity varied from 0.000 to 0.8929 and from 0.000 to 0.7662, respectively.

本研究针对雷曼普亚凤梨(Puya raimondii)、大穗普亚凤梨(P. macrura)与哈钦森普亚凤梨(P. hutchinsonii)开展基因组浅层测序(genome skimming),筛选并合成70对跨物种简单重复序列(Simple Sequence Repeats, SSR)引物。其中70对引物中有42对可成功实现跨物种扩增。经初步筛选后,12对SSR引物能够扩增出清晰条带,遂被用于检测采自秘鲁4个种群的84株雷曼普亚凤梨个体的潜在多态性。结果显示,每个位点的等位基因数量介于1至6之间,观测杂合度与期望杂合度的取值范围分别为0.000~0.8929和0.000~0.7662。
