We report the transcriptome changes in purified mouse muscle stem cells in response to deletion of the Cox10 gene. This study identifies changes in the muscle stem cell population in response to mitochondrial dysfunction. Overall design: A floxed-Cox10 allele combined with Pax7-Cre (ERT2) driver were used to induce Cox10 deletion in muscle stem cells of adult animals, via administration of tamoxifen. Two days after tamoxifen administration, muscle stem cells were purified from wild-type (control) and Cox10-/- animals.

本研究报道了纯化小鼠肌肉干细胞（muscle stem cell）中，Cox10基因敲除所引发的转录组（transcriptome）变化。本研究明确了线粒体功能障碍刺激下，肌肉干细胞群体所发生的转录组改变。
整体实验设计：借助他莫昔芬给药，通过搭载Cox10条件性敲除等位基因（floxed-Cox10 allele）与Pax7-Cre（ERT2）驱动系统的体系，在成年动物的肌肉干细胞中诱导Cox10基因敲除。在他莫昔芬给药两天后，分别从野生型（对照组）与Cox10基因敲除（Cox10-/-）动物体内纯化得到肌肉干细胞。
