Dying pancreatic cancer cells from chemoradiation can directly stimulate repopulation of surviving cells. We used microarrays to conduct the analysis of whole transcript profiles in pancreatic cancer cells treated with X-ray radiation. Pancreatic cancer cells were seeded into culture plate overnight with 2% FBS in culture medium and then were irradiated with 10Gy using an Oncor linear accelerator. Irradiated pancreatic cancer cells were collected at 24h after X-ray radiation for RNA extraction and hybridization on Affymetrix microarray. We sought to obtain whole transcript profiles in pancreatic cancer cells treated with X-ray radiation.
经放化疗后濒死的胰腺癌细胞可直接刺激存活肿瘤细胞的再增殖。我们采用微阵列(microarrays)技术,对经X射线辐射处理的胰腺癌细胞的全转录组谱展开分析。将胰腺癌细胞以含2%胎牛血清(Fetal Bovine Serum, FBS)的培养液接种于培养板中过夜培养,随后使用Oncor直线加速器以10Gy剂量进行X射线辐射。于X射线辐射后24小时收集受辐射的胰腺癌细胞,进行RNA提取并在Affymetrix微阵列上完成杂交反应。本研究旨在获取经X射线辐射处理的胰腺癌细胞的全转录组表达谱。
