14 different AAV vectors were packaged with 14 barcoded AAV transgenes. NGS was used to de-multiplex the composition of the barcoded mix at the vector level, at the entry level (DNA) and at the expression level (cDNA) of the targeted tissues, which were human liver ex plants, murine liver, xenografted murine liver and non-human primate liver.
本研究将14种不同的腺相关病毒载体(AAV vectors)分别与14个带有条形码的腺相关病毒转基因元件进行包装。采用下一代测序(NGS, Next Generation Sequencing)技术,对靶向组织的载体层面、输入水平(DNA层面)以及表达水平(cDNA层面)的条形码混样组成进行解多重分析;所涉及的靶向组织包括人类肝脏外植体、小鼠肝脏、异种移植小鼠肝脏以及非人类灵长类肝脏。
