High throughput sequencing was used to investigate the production of small RNAs from in Chlamydomonas in different strains and different stages of the life cycle. The association between these and methylation was assessed using genomic sequencing, comparing a sequenced genome with one enriched for methylated DNA sequence by immuno-precipitation. Overall design: Examination of small RNA production in several strains and of the methylation of regions producing these small RNAs.
本研究采用高通量测序(High-throughput sequencing)技术,探究衣藻(Chlamydomonas)不同品系及生命周期不同阶段的小RNA(small RNAs)产生情况。通过基因组测序(genomic sequencing)评估上述小RNA与甲基化的关联:将常规测序获得的基因组与经免疫沉淀(immuno-precipitation)富集的甲基化DNA序列基因组进行对比分析。整体实验设计:对多个品系的小RNA产生情况,以及产生这些小RNA的基因组区域的甲基化水平开展检测。
