RNAseq analysis to identify differential expressed genes in glomerular cDNA of diabetic DBA/2 mice with Esm1 over-expression

We aim to study the gene expression changes in glomeruli of DBA/2 mice with diabetic nephropathy with or without Esm-1 over-expression. We extract pure mice glomeruli, prepared total cDNA with random hexamers and performed RNAseq study to quantify transcriptome and identify differential expressed genes. With a sequencing depth of 20M, we detected 12934 genes in total. We found many interferon related genes shows differences between control and DM group, suggesting DBA/2 mice with diabetic nephropathy are driven by interferon related mechanisms. We found 41 genes show biologically meaningful differences between DM and Esm-1 + DM group, suggesting these genes are downstream targets of Esm-1 in mice. Quantify glomerular transcriptome changes with Illumina 1.9 in 10 weeks diabetes mice with or without Esm-1 over-expression.
本研究旨在探究过表达或未过表达内皮细胞特异性分子1(Esm-1)的糖尿病肾病DBA/2小鼠肾小球内的基因表达变化。我们提取纯化的小鼠肾小球,采用随机六聚体引物制备总cDNA,随后开展RNA测序(RNAseq)以定量转录组并鉴定差异表达基因。本研究的测序深度为20M,共检测到12934个基因。我们发现对照组与糖尿病(DM)组间存在大量干扰素相关基因的表达差异,提示糖尿病肾病DBA/2小鼠的发病机制与干扰素通路相关。本研究还发现糖尿病组与Esm-1过表达糖尿病组间存在41个具有生物学意义的差异表达基因,提示这些基因是小鼠体内Esm-1的下游靶标。本研究采用Illumina 1.9测序平台,对造模10周的过表达或未过表达Esm-1的糖尿病小鼠肾小球进行转录组变化定量分析。
