Lactobacillus NK318.1 is a commensal microbe isolated from the mouse intestinal feces in our lab. To examined the potential role of L. NK318.1 in the gut immunity, we monocolonized GF mice with L.NK318.1. And, we conducted a microarray experiment to compare the transcriptomes of GF and NK318.1 mice intestines under the same experimental condition We used microarrays to detail the global programme of gene expression in intestinal epithelial cells (IEC) and Peyer's patches cells (PP) of GF and NK318.1 mice. Overall design: IEC and PP of GF and NK318.1 mice of the same physiological and maintaining condition were isolated and then total RNAs were extracted from them, which could reflect the respective transcription profile of GF and NK318.1 mice. These RNAs were detected by microarray and the influence of L.NK318.1 on intestine could be revealed to some extent.

乳杆菌NK318.1是本实验室从小鼠肠道粪便中分离得到的共生微生物。为探究乳杆菌NK318.1在肠道免疫中的潜在作用,我们采用该菌株对无菌(GF)小鼠进行单菌定植。随后,我们在相同实验条件下开展基因芯片实验,以比较无菌小鼠与定植乳杆菌NK318.1小鼠的肠道转录组差异。本研究利用基因芯片解析无菌小鼠与定植乳杆菌NK318.1小鼠的肠上皮细胞(IEC)及派尔集合淋巴结细胞(PP)的全基因表达谱。实验设计:将饲养条件与生理状态一致的无菌小鼠及定植乳杆菌NK318.1小鼠的肠上皮细胞与派尔集合淋巴结细胞分离,随后提取总RNA,以此反映两类小鼠各自的转录组特征。通过基因芯片对上述总RNA进行检测,可在一定程度上揭示乳杆菌NK318.1对肠道的影响。
