Data from Aquaref 2024 project, metabarcoding for the Diatoms Biological Index, France

These data were generated as part of the Aquaref 2024 project for the assessment of the Diatom Biological Index (DBI) required by the Water Framework Directive (WFD). Samples were collected by french Water Agencies and OFB laboratories (Adour-Garonne, Rhin-Meuse, Loire-Bretagne, Rhône-Méditerannée-Corse, Seine-Normandie, Artois-Picardie). DNA was extracted using the NucleoMag® DNA Microbiome kit (Macherey-Nagel). DNA was amplified by PCR using RbcL primers (3FW + 2RV). FastQ files were obtain after DNA sequencing based on the Illumina MiSeq technology with the v3 reagent kit (2 x 250pb) at the GeT-Biopuces platform (Toulouse, France). This directory contains the following files : Aquaref_2024_Run1.tar.gz : all samples FastQ files from the first run 2024 (368 x 2 replicates) Aquaref_2024_Run2.tar.gz : all samples FastQ files from the second run 2024 (57 x 2 replicates) Metadata_Aquaref_2024.xlsx : contains all the data available for each samples corresponding to the names of FastQ files. Metadata_Aquaref_2024.csv : contains all the data available for each samples corresponding to the names of FastQ files.本数据集系Aquaref 2024项目的研究产出，用于评估《水框架指令》(Water Framework Directive, WFD)所要求的硅藻生物指数(Diatom Biological Index, DBI)。样本由法国水务机构及法国生物多样性办公室（OFB）实验室采集，采样覆盖阿杜尔-加龙、莱茵-默兹、卢瓦尔-布列塔尼、罗讷-地中海-科西嘉、塞纳-诺曼底以及阿尔图瓦-皮卡第六大流域区域。实验采用NucleoMag® DNA微生物组试剂盒（Macherey-Nagel品牌）完成基因组DNA提取，以RbcL引物（3FW + 2RV）通过聚合酶链式反应（Polymerase Chain Reaction, PCR）完成DNA扩增。依托法国图卢兹的GeT-Biopuces测序平台，采用Illumina MiSeq测序技术搭配v3试剂试剂盒（2×250碱基对）完成高通量测序，最终得到FastQ格式文件。 本数据集目录包含以下文件： Aquaref_2024_Run1.tar.gz：收录2024年首轮测序的全部样本FastQ格式文件（共368组，每组含2个重复样本） Aquaref_2024_Run2.tar.gz：收录2024年次轮测序的全部样本FastQ格式文件（共57组，每组含2个重复样本） Metadata_Aquaref_2024.xlsx：包含所有与FastQ文件名称对应的样本元数据信息 Metadata_Aquaref_2024.csv：包含所有与FastQ文件名称对应的样本元数据信息
