发现了靶向TRADD可以同时恢复细胞内稳态并抑制细胞凋亡。采用RIPK1依赖的细胞死亡模型和细胞自噬模型，从17万个化合物中进行了多轮筛选，最终获得了活性小分子Apostatin-1并证明了其作用靶点为TRADD蛋白。通过体外和体内实验进一步证明了靶向TRADD可以实现同时抑制RIPK1的活化介导的细胞凋亡和激活自噬恢复细胞内稳态并清除病理蛋白聚集物
It was discovered that targeting TRADD can simultaneously restore intracellular homeostasis and inhibit apoptosis. Using RIPK1-dependent cell death models and autophagy models, multiple rounds of screening were conducted against 170,000 compounds, and the active small molecule Apostatin-1 was finally isolated, with its target protein verified to be TRADD. Further validation through in vitro and in vivo experiments confirmed that targeting TRADD can both inhibit RIPK1 activation-mediated apoptosis and activate autophagy to restore intracellular homeostasis and clear pathological protein aggregates.
