2-week old Arabidopsis thaiana that grew on the liquid root culture were infected with Verticillium dahliae JR2 strain. The infected samples were collected at 2 and 4 days post inoculation, respectively. These samples were equally mixed, then process to Arabidopsis AGO1 or AGO2 immunoprecipitation (IP), AGO1 or AGO2 associated RNAs were used to construct small RNA library following the miRCat small RNA cloning system (IDT, Coralville, IA).
将在液体根培体系中生长的2周龄拟南芥(Arabidopsis thaliana)接种大丽轮枝菌(Verticillium dahliae)JR2菌株,分别于接种后2天和4天采集感染样本。将采集的样本等量混匀后,开展拟南芥AGO1或AGO2的免疫沉淀(immunoprecipitation, IP)实验;以AGO1或AGO2结合的RNA为材料,参照miRCat小RNA克隆体系(IDT,美国爱荷华州科勒尔维尔市)构建小RNA文库。
