To analyse the genome-wide impact of inactivation of the Supt3h subunit of the SAGA complex on RNA polymerase II (Pol II) transcription in human U2OS cells and mouse embryonic stem cells (mESCs), we analysed newly synthesized RNA using 4sU-labelling in mutant and wildtype conditions. Newly synthesized RNA from wildtype (WT) and mutant conditions of human U2OS cells and mESCs. mESCs were cultured in FCS+2i conditions. Samples were sequenced in two independent sequencing projects and RNA from two independent clones per cell line were sequenced.

为分析SAGA复合物(SAGA complex)的Supt3h亚基(Supt3h subunit)失活对人类U2OS细胞与小鼠胚胎干细胞(mouse embryonic stem cells,mESCs)中RNA聚合酶II(RNA polymerase II,Pol II)转录的全基因组影响,本研究针对人类U2OS细胞及mESCs在野生型(wildtype,WT)与突变体条件下的新合成RNA,采用4sU标记法(4sU-labelling)开展分析。其中mESCs采用FCS+2i培养体系进行培养,所有样本均通过两个独立测序项目完成测序,且每个细胞系的两份独立克隆的RNA均被测序。
