Systems pharmacology demonstrates broad-spectrum efficacy in animal models of retinopathies [scRNA-seq: Rpe65-KO]

To determine the gene expression changes in the retina P57-P60 Rpe65-/- mice , we collected retina samples from Rpe65-/- and WT (C57BL/6J mice) mice, generated single-cell suspensions, and performed scRNA-Seq (10X Genomics). One group of Rpe65-/- mice was treated for 5 weeks via dietary administration of tamsulosin, metoprolol and bromocriptine drug (TMB) cocktail. Vehicle- and TMB-treated Rpe65-/- mice: One retina from seven male Rpe65-/- mice were pooled to prepare single-cell suspensions at a final concentration of 1,000 cells/l. WT mice: One retina from three age-matched male C57BL/6J mice were pooled to prepare single-cell suspensions at a final concentration of 1,000 cells/l. For each group, freshly dissociated cells (~10,000) were loaded into a 10x Genomics Chromium Single Cell system using v3 chemistry following the manufacturer's instructions.
