TFAP2A (AP-2α) is a member of the transcription factor AP-2 family, functions as a tumor suppressor by regulating the expression of various cancer-related genes; its expression is positively correlated with chemosensitivity and survival of cancer patients. Overall design: UM-UC-3 cells were infected with a lentivirus expressing empty vector or AP-2α cDNA for 48 h and then the cells were selected with puromycin (1 µg/mL). Cells infected with empty vector (control) and AP-2α cDNA were sequenced by Illumina HiSeq2000.

TFAP2A(AP-2α)属于转录因子AP-2家族成员,可作为肿瘤抑制因子,通过调控多种癌症相关基因的表达发挥生物学功能;其表达水平与癌症患者的化疗敏感性及生存率呈正相关。实验整体设计:将UM-UC-3细胞感染携带空载体或AP-2α互补DNA(cDNA)的慢病毒,培养48小时后,使用浓度为1 µg/mL的嘌呤霉素进行细胞筛选。分别对空载体感染组(对照组)与AP-2α cDNA感染组的细胞开展Illumina HiSeq2000测序。
