The study objective was to determine differentially expressed mRNA transcripts in cardiac tissues from E18.5 fetal mice e born to obese dams fed a high fat/high sugar diet and control dams fed normal diet. Overall design: Twelve-week-old C57BL/6J female mice were fed either a control diet (D12489, 10% calories from fat) or an obesogenic diet (Western Diet D12089B; Research Diets, 40% of calories from fat). Female mice fed the obesogenic diet had ad libitum access to 20% sucrose solution supplemented with micronutrients. Females were mated following a 25% gain in weight over the initial body weight in the obesogenic diet. All males were fed control diet. At E18.5, animals were euthanized and hearts were flash frozen. Following RNA isolation, RNAseq was performed.
本研究旨在明确喂食高脂高糖饲料的肥胖母鼠与正常饮食对照组母鼠所产的E18.5胎鼠心脏组织中,差异表达的信使核糖核酸(mRNA)转录本。实验设计概述:选取12周龄的C57BL/6J雌性小鼠,分别喂食对照饲料(D12489,热量占比10%来自脂肪)或致肥胖饲料(Western Diet D12089B;Research Diets出品,热量占比40%来自脂肪)。喂食致肥胖饲料的雌性小鼠可自由采食添加了微量营养素的20%蔗糖溶液。待致肥胖饲料组小鼠体重较初始体重增加25%后,将其与雄性小鼠合笼交配,所有雄性小鼠均喂食对照饲料。于胚胎发育第18.5天(E18.5)处死实验动物,采集心脏组织并快速冷冻。提取RNA后,进行RNA测序(RNAseq)。
