本实验揭示了转导非靶向单引导RNA(non-targeting single-guide RNA, NT-sgRNA)或靶向Bap1基因启动子的单引导RNA(sgRNA1)的小鼠滋养层干细胞(mouse trophoblast stem cells)在两种时间节点的转录组变化:0天(干细胞维持培养条件,即未分化状态)与诱导分化3天(3d)后。 实验整体设计:本实验通过设置三次生物学重复的RNA测序(RNA-seq),在0天和诱导分化3天两个时间点,对比对照组(转导NT-sgRNA)与转导sgRNA1的Bap1过表达小鼠滋养层干细胞(mTSCs)的转录组差异。
