RNAseq analysis of DDX21-silenced colorectal cancer cell HCT8

DEAD-box RNA helicase 21 (DDX21), is a nucleolar protein harboring ATP-dependent double-stranded RNA unwinding activities, essential in rRNA processing and ribosome biogenesis. However, its role in colorectal cancer (CRC) progression remains unclear. In this study, we performed RNAseq in colorectal cancer line HCT8 with or without DDX21 gene silencing, to reveal the role of DDX21 in transcriptional and epigenetic control of CRC cell proliferation. DEAD盒RNA解旋酶21（DEAD-box RNA helicase 21, DDX21）是一种核仁蛋白，具备ATP依赖型双链RNA解旋活性，在核糖体RNA加工与核糖体生物发生过程中不可或缺。然而，其在结直肠癌（colorectal cancer, CRC）进展中的作用仍不明晰。本研究对存在或缺失DDX21基因沉默的结直肠癌HCT8细胞系开展RNA测序（RNAseq），以揭示DDX21在结直肠癌细胞增殖的转录与表观遗传调控中的作用。总体实验设计：从经DDX21基因沉默与未沉默的HCT8细胞中分离总RNA，随后进行RNA测序。
