Quantitative regulation of Arabidopsis FLC involves a transcription-coupled chromatin mechanism whereby the decision to polyadenylate at proximal or distal sites is linked to histone demethylation of adjacent chromatin. To analyse the polyadenylation site choice in an unbiased and strand-specific manner, we carried out Quant-seq analysis on total RNA from 7DAG seedlings in Col-0, fca-9, and fld-4. To increase coverage fld4 libraries were enriched using an in solution bait target capture approach.
